Prognostic Value of Apurinic Endonuclease (APE) Expression in Bladder Cancer

Abstract
Purpose: Oxidative stress is involved in many types of DNA damage which have been implicated in mutagenesis and considered to play an important role in human cancer development. This DNA damage is recognized and repaired by variable gene repair systems such as apurinic endonuclease (APE). In this study, APE was analyzed by case-control study to investigate their association with bladder cancer and possible interaction with smoking status.

Materials and Methods: To determine the expression of APE, mRNAs extracted from 100 bladder cancer tissues and 18 normal bladder mucosae were analyzed using quantitative competitive PCR (QC-PCR). Smoking status was obtained from self-completed questionnaire. The expression of APE was compared in each group and its difference according to bladder cancer risk factors including pathologic stage, grade, recurrence, and smoking status were also analyzed in bladder cancer group.

Results: The expression of APE was not different between the bladder cancer and the normal bladder tissues. The expression of APE in primary bladder cancer tissues was significantly higher in non-recurred group than recurred cases (0.86¡¾0.93 vs 0.45¡¾0.66, respectively) (p=0.031). Among the bladder cancer group, nonsmokers` tissues expressed significantly higher level of APE than of smokers` (1.02¡¾1.17 vs 0.60¡¾0.70) (p=0.049). The expression of APE did not vary with pathologic stage and grade.

Conclusions: The expression of APE in recurred bladder cancer tissues and smokers was remarkably decreased compared with in the non-recurred cases and nonsmokers. These results suggest that APE expression may be altered by environmental factors such as smoking and might be a useful prognostic marker for bladder cancer recurrence.
